Sam wears a LOT of plaid shirts, so I'd say that no, there's no pattern with him and plaid.
Dean on the other hand only has a few plaid shirts. I always associated Dean and plaid with more domestic scenes...fixing the Impala, when he was in high school, when he's in the heaven memory with his mom...but there are instances that sort of break that: He wears plaid during both shapeshifter episodes. He also wears plaid during Time is On My Side - which is a tough episode emotionally, because his time is running out and he and Sam are at odds as to what they should be doing.
But, yeah, if there's any pattern with plaid - I'd say it's that Dean wears it when he's more domestic...whether that means he's happy or not is a matter of character interpretation.
